THE FORMULA — 11 BIOACTIVES. FOUR LONGEVITY PILLARS.

PILLAR ONE: AUTOPHAGY ACTIVATION

Puremidine® (Spermidine) — 10 mg

Role: Master Autophagy Activator, Polyamine Longevity Signaling & Cognitive Resilience

Spermidine is a naturally occurring polyamine found in all living cells — and one of the most important endogenous regulators of autophagy, the cellular recycling and quality-control process that clears damaged proteins, dysfunctional organelles, and cellular debris.* Spermidine levels decline progressively with age in virtually every tissue, including the brain — and this decline is increasingly recognized as a primary driver of the accumulation of cellular damage that characterizes aging.*

Puremidine® is a pure synthetic spermidine — GRAS-approved, allergen-free, and free from the contaminants that can affect wheat-derived spermidine sources. At 10 mg, Prime Time+ delivers three times the 3.3 mg dose used in published clinical trials demonstrating cognitive benefits — the highest-dose daily supplemental spermidine formula in the Healthgevity catalog.*

Published Clinical and Research Evidence:*

Human cognitive trial: Clinical trials in elderly adults with mild cognitive impairment demonstrated that 3.3 mg daily spermidine significantly improved cognitive test scores — with high-dose groups showing approximately 50% greater gains than low-dose groups, including improvements in verbal fluency, memory, and overall cognitive function.*

2025 human clinical study (Aging journal): Spermidine supplementation significantly reversed age-associated memory decline — while protein restriction alone (another longevity intervention) failed to protect against memory decline. These two approaches work through independent mechanisms, with spermidine protecting cognitive function through pathways protein restriction cannot access.*

2025 Randomized Clinical Trial (POLYCAD, Denmark): Spermidine's effects in elderly cardiac patients are under investigation in a completed enrollment RCT examining cardiovascular aging, left ventricular function, and physical performance (NCT06186102, results anticipated 2026).*

2025 double-blind, randomized, placebo-controlled study in adults over 65: Spermidine reduced immune cell senescence (decreased p16 expression in lymphocytes), enhanced autophagy, and supported vaccine immune response in older adults — demonstrating both autophagy activation and immune senescence mitigation.*

2026 Nature review: Identified autophagy activation as the most strongly supported mechanism behind spermidine's longevity effects — supporting mitochondrial function, stress resistance, and epigenetic regulation alongside cellular recycling.*

Key biological mechanisms:*

• Autophagy induction through inhibition of acetyltransferases and histone deacetylation modulation*
• Hypusination of eIF5A — a translational factor required for mitochondrial integrity and memory function in aging brain*
• Mitochondrial protection and improved mitochondrial respiration*
• Anti-inflammatory signaling — spermine (spermidine's downstream polyamine) inhibits pro-inflammatory cytokine synthesis*
• Epigenetic regulation of longevity-associated gene expression*

2024 Nature Cell Biology study: Spermidine is essential for fasting-mediated autophagy and longevity — meaning many of the cellular benefits of caloric restriction and intermittent fasting are mediated through spermidine's induction of autophagy. Prime Time+ delivers these benefits without requiring caloric restriction.*

Fucoidan (Mekabu / Undaria pinnatifida Sporophyll Extract, standardized to 85% fucoidan)

Role: Autophagy Support, Glycocalyx Integrity & Stem Cell Mobilization Support Dose: Standardized 85% fucoidan

Fucoidan is a sulfated polysaccharide extracted from the sporophyll of Undaria pinnatifida — the same Mekabu fucoidan featured in Cardio NAD+ for its glycocalyx support. In Prime Time+, fucoidan contributes to autophagy regulation, vascular glycocalyx maintenance, and stem cell mobilization support.*

Emerging research demonstrates fucoidan's role in autophagy pathway regulation — promoting autophagic flux in a complementary mechanism to spermidine's polyamine-mediated induction.*

Fucoidan also supports the vascular glycocalyx — the protective carbohydrate layer coating the endothelial surface of blood vessels — which is critical for circulatory health, microvascular flow regulation, and inflammatory tone in the vascular compartment.*

Luteolin & Apigenin

Role: NAD+ Conservation, Autophagy Support & Neuro-Vascular Defense

Luteolin and Apigenin are two complementary dietary flavonoids with overlapping and synergistic roles in cellular longevity support.

Apigenin supports NAD+ homeostasis by modulating CD38 — a primary enzyme responsible for NAD+ degradation. By helping conserve endogenous NAD+, apigenin supports the sirtuin and PARP activity that depends on NAD+ availability, contributing to epigenetic longevity signaling alongside spermidine's autophagy activation.*

Luteolin provides complementary antioxidant defense, neuro-vascular resilience support, and healthy inflammatory response modulation — contributing to the cellular environment in which autophagy and renewal processes can operate most effectively.*

Both flavonoids also support autophagy signaling independently — apigenin through mTOR pathway modulation, luteolin through oxidative stress reduction — reinforcing spermidine's autophagy activation through distinct and complementary molecular mechanisms.*

PureMune™ (Yeast β-1,3/1,6 Glucan)

Role: Innate Immune Priming & Immune-Linked Autophagy Activation

PureMune™ is a patented, highly active beta-glucan fraction derived from yeast cell walls, formulated for potent innate immune activation. Beta-glucans are well-characterized pattern recognition ligands that interact with the Dectin-1 receptor on macrophages, dendritic cells, and NK cells — priming innate immune surveillance and balanced inflammatory response.*

In Prime Time+, PureMune™ serves a dual role: direct innate immune activation, and contribution to immune-linked autophagy. Autophagy and immune function are deeply interconnected — macrophage-mediated autophagy is a primary mechanism for clearing senescent cells and cellular debris, and PureMune™'s macrophage activation supports this immune-autophagy crosstalk.*

PILLAR TWO: CELLULAR RENEWAL & STEM CELL MOBILIZATION SUPPORT

StemEnhance® (Aphanizomenon flos-aquae Extract)

Role: Bone Marrow-Derived Circulating Stem Cell Mobilization Support

StemEnhance® is a patented water-soluble extract of Aphanizomenon flos-aquae (AFA) — a freshwater blue-green algae from Klamath Lake — standardized for the active fractions shown to support the natural release of bone marrow-derived stem cells into circulation.*

The bone marrow contains a reservoir of adult stem cells — including hematopoietic stem cells (HSCs), characterized by the CD34+ surface marker — that circulate in the bloodstream and migrate to sites of tissue wear to participate in repair and maintenance processes. The number of circulating CD34+ stem cells naturally declines with age, and is recognized as a meaningful parameter for the body's regenerative capacity.*

StemEnhance® has been shown in human studies to support the mobilization of these cells:*

• Double-blind study in 22 healthy adults: AFA extract consumption significantly increased the number of circulating peripheral CD34+ cells within 48 hours. Long-term administration maintained elevated CD34+ cell levels at 7-day and 38-day follow-up.*
• Crossover, double-blinded, placebo-controlled study (21 volunteers): AFA consumption produced generalized mobilization of lymphocytes and monocytes within 2 hours, including increases in CD3+, CD4+, and CD8+ T cell subsets and CD19+ B cells — suggesting broad immune-cell mobilization activity.*
• Animal model study: StemEnhance significantly improved muscle regeneration following cardiotoxin-induced injury, associated with enhanced bone marrow stem cell mobilization and migration to sites of tissue damage.*

StemEnhance® also provides phycocyanin, phosphatidylserine, and phenylethylamine (PEA) — contributing to the formula's immune modulation, cellular defense, and mood balance dimensions.*

Important framing: the mobilization of bone marrow-derived circulating stem cells into the peripheral bloodstream is a normal physiological process that supports tissue maintenance and repair. StemEnhance® supports this natural process. Prime Time+ does not contain stem cells and does not claim to regenerate tissue through stem cell therapy.*

CyanthOx™ (Sea Buckthorn / Hippophae rhamnoides Berry Extract)

Role: Endothelial & Vascular Longevity, Antioxidant & Stem Cell Environment Support

CyanthOx™ is derived from wild Sea Buckthorn from the Tibetan Plateau — one of the most polyphenol-dense botanical sources available — providing a synergistic complex of proanthocyanidins, flavonols, carotenoids, and hydroxycinnamic acids with broad antioxidant and vascular-protective activity.*

Clinical evidence for CyanthOx™ demonstrates:*

• Support for endothelial function, vascular elasticity, and healthy blood flow*
• Antioxidant protection against lipid peroxidation in vascular tissue*
• Support for healthy skin integrity and collagen-associated tissue health*
• Balanced inflammatory signaling in the vascular compartment*

In the context of Prime Time+'s stem cell mobilization support, healthy vascular endothelial function is directly relevant — mobilized circulating stem cells must migrate through vascular endothelium to reach sites of tissue maintenance. CyanthOx™ supports the endothelial environment that facilitates this process.*

Alomac® (Aloe macroclada Leaf Gel Extract)

Role: Cellular Resilience, Epithelial Integrity & Regenerative Signaling Support — NEW

Alomac® is a novel ingredient derived from Aloe macroclada — a rare aloe species found exclusively in Madagascar, distinct from the common Aloe vera used in most aloe supplements. Aloe macroclada provides unique polysaccharides and bioactive compounds not found in conventional aloe extracts.*

Research on Aloe macroclada-derived extracts demonstrates:*

• Support for cellular resilience and epithelial barrier integrity*
• Balanced inflammatory response signaling through natural biological pathways*
• Support for tissue maintenance and healthy cellular regeneration signaling*
• Antioxidant and cytoprotective activity*

Alomac® represents the kind of novel botanical innovation that distinguishes Prime Time+ from conventional longevity formulas — an ingredient with a biologically compelling profile and meaningful differentiation from commodity botanical extracts.*

Astaxanthin

Role: Membrane-Integrated Antioxidant, Mitochondrial Protection & Cellular Aging Defense

Astaxanthin in Prime Time+ serves dual roles across Pillar 2 (cellular renewal) and Pillar 3 (mitochondrial resilience). Its unique membrane-spanning structure — with polar end groups bridging both sides of the lipid bilayer simultaneously — provides antioxidant protection across the full membrane thickness, protecting cell membranes and mitochondrial membranes from lipid peroxidation.*

Astaxanthin also supports autophagy signaling, stem cell function, and cellular renewal processes — contributing to Pillar 2 alongside StemEnhance® and CyanthOx™.*

At the higher dose in Prime Time+, astaxanthin is in the upper range of doses used in human clinical trials for oxidative stress, cognitive performance, and exercise recovery.*

PILLAR THREE: MITOCHONDRIAL & METABOLIC RESILIENCE

Alpha-Ketoglutarate (AKG)

Role: Krebs Cycle Intermediate, Epigenetic Longevity Regulation & Metabolic Resilience

Alpha-ketoglutarate is a key metabolic intermediate in the Krebs cycle — the central energy-generating pathway in all aerobic cells. AKG serves as a co-substrate for over 70 enzymes in human biology, including the TET enzymes and Jumonji-domain demethylases that regulate DNA methylation and histone modification — epigenetic mechanisms increasingly linked to the hallmarks of aging.*

Published research demonstrates AKG's role in:*

• Supporting healthy mitochondrial energy production and amino acid synthesis*
• Modulating the epigenetic clock — DNA methylation patterns associated with biological aging*
• Promoting cellular detoxification through the urea cycle and transamination*
• Supporting healthy mTOR regulation and balanced metabolic signaling*
• Contributing to collagen and connective tissue synthesis*

AKG is particularly relevant in the context of the intermittent fasting and caloric restriction longevity literature — fasting increases endogenous AKG availability, and supplemental AKG in longevity contexts provides fasting-related metabolic benefits without requiring caloric restriction.*

CyaninPlus® (Blue Spirulina Extract, ≥30% Phycocyanin)

Role: COX-2 Modulation, Cellular Energy Support & Antioxidant Neuroprotection

CyaninPlus® provides concentrated phycocyanin — the blue photosynthetic pigment of spirulina and one of the most potent natural COX-2 pathway modulators identified in plant biology.* Phycocyanin's anti-inflammatory activity is structurally mediated through its tetrapyrrole chromophore, which scavenges peroxyl radicals and inhibits inflammatory enzyme activity directly.*

Phycocyanin also:*

• Supports liver detoxification and antioxidant enzyme activity*
• Contributes to healthy immune function and NK cell activity*
• Provides cellular energy support through photosynthetic pigment-derived electron-transfer activity*
• Supports neurological antioxidant defense — phycocyanin crosses the blood-brain barrier*

PILLAR FOUR: IMMUNE BALANCE & INFLAMMATORY MODULATION

(PureMune™, CyaninPlus®, StemEnhance® AFA, Fucoidan — all described above, contributing across pillars)
